Study details
Doctors enrolled 320 subjects who had used AZT for at least 6 months but who had never taken protease inhibitors or 3TC. At the start of the study 50% of subjects had a CD4+ count of 15 cells and a viral load of 91,210 copies. Subjects received indinavir 2.4 g/day or a combination of AZT/3TC or all three drugs (triple therapy) at standard doses.
Results
By the 6th month of the study, subjects on ?triple therapy? had the greatest decrease in viral load. Indeed, 65% of them had their viral load fall to below 500 copies. Subjects on triple therapy had their CD4+ count rise by about 86 cells, also at the 6th month.
Indinavir with nevirapine and 3TC
Doctors in Vancouver recruited 21 subjects (2 female, 19 male) who had less than 51 CD4+ cells and had extensively used anti-HIV drugs and experienced serious toxicity with them. Subjects received indinavir 2.4 g/day, nevirapine 200 mg/day for the first 2 weeks, after which it was given at a dose of 400 mg/day; and 3TC 300 mg/day.
Results
By the 8th week, 10 subjects who had entered the study with a viral load of 190,546 copies, had their viral load fall to 851 copies. The abstract did not have information on CD4+ cell counts. It will be interesting to see if this decrease in viral load continues and is linked with beneficial effects. Although no subject developed kidney stones, two had intestinal problems because of indinavir and had to leave the study. At least one person had a skin rash from nevirapine.
