The dance music star, who has now returned to Hove, Sussex, said: "If we've saved one life, it's worth it."
Cook, famous for his love of Brighton and Hove Albion FC, was the patron of Coaching for Hope, run by development agency International Service.
Football coaches from the Brighton team worked with the children in Mali.
And HIV workshops included art activities and discussions.
Cook said: "Football always brings people together in the way that music does, which is my business.
"It is fun for the kids but also there's something else attached to it that is far more important."
He said: "If it saves one life it's worth doing it.
And he added: "I was just astounded by my lack of talent at art, matched only by my lack of talent at football.
"I think when I get home I'll stick to music."
The development agency's scheme is also being run in Latin America and the Middle East.
