It will be the first time so many top stars have joined together for charity since Band Aid in the 1980s.
Britney Spears, U2's Bono and Destiny's Child are among the artists who will record a cover version of Marvin Gaye's classic What's Going On.
The song will be released in America on 1 December - World Aids Day.
Bono took on the task of bringing the American stars together after a request from British Aids campaigner Leigh Blake.
Proceeds
The aim is not only to raise awareness of the plight of Aids victims in the African continent but also to persuade world leaders to do more.
Proceeds of the single will go towards Aids programmes and research by organisations working in Africa.
Speaking on BBC News 24 Bono said: "We can be remembered for the internet and the time when the world woke up to its responsibilities to its neighbours."
And in USA Today he said he would be happy for a younger pop star to take his place.
"Part of the energy to this is getting a new mix of people."
Other artists taking part include Mary J Blige, Wyclef Jean and rapper Ja Rule.
Developing countries
The song is being produced by Jermaine Dupri, who has worked with Mariah Carey and Janet Jackson.
As many as 95% of the 36 million people who are living with HIV or Aids live in the developing countries.
And in parts of Africa someone dies from the disease every 13 seconds.
Those helping the plight of the victims say the support from the rock industry is very significant.
Although the single is only due to be released in America the organisers are considering whether it should widen the release to Britain.
