"Trials of Colony Stimulating Factors Grow, So Do Applications, Side" Effects

Abstract: A wealth of leukocytic riches may result from the use of colony stimulating factors (CSF) to amplify celluar immune response, but early clinical trials also show that toxic effects are beginning to appear at high-dose levels. One CSF, granulocyte-macrophage colony stimulating factor (GM-CSF), has been tested on patients with AIDS as well as those with several other diseases. A University of California at Los Angeles study involving patients with AIDS showed that GM-GSF the recombinant substance led to a dose-dependent increase in circulating leukocytes, especially neutrophils. At lower doses, bone pain was the main side effect. When the dose was raised slightly, the side effects included pericarditis, pleuritis, and eosinophilia with pulmonary infiltrates in patients with aplastic anemia at UCLA. Granulocyte colony stimulating factor (G-CSF) may be less toxic, however. Questions remain about whether increased white cells resulting from CSFs reduce infections and whether the substances will allow for the escalated use of chemotherapy. Researchers need more research to find answers to these and other questions.
