PHYLOGENETIC ANALYSIS OF HUMAN IMMUNODEFICIENCY VIRUS TYPE 1 CRF07_BC PREVAILING IN THE TWO MAJOR PROVINCES IN CHINA

BACKGROUND: HIV-1 CRF07_BC was first reported in China in 1996, and spread along heroin trafficking routes from Southwest to northwest of China. The original transmission of the virus has not yet been elucidated.
METHODS: 193 HIV-1 env sequences (V3 to V4) generated between 1997 and 2005 from IDUs in Lishang (63), Urumqi (57)and Yili (73), with 5-7 sampling time points. All the sequences were genotyped as HIV-1 CRF07_BC, according to the Neighbor-Joining tree with Mega4.0. The sequences were categorized based on geographic and demographic information, then analyzed with Neighbor-joining tree and Maximum likelihood tree by PAUP4b10. The major clusters were selected for tMRCA estimate using Bayesian evolutionary analysis sampling trees (BEAST1.4.6).
RESULTS: Phylogenetic analysis indicated that the transmission and spread of CRF07_BC happened after immigration into the local regions. 44 out of 63 sequences in Liangshan, 49 in 57 from Urumqi, 58 in 73 from Yili formed the major monophylogenetic clusters, respectively. The data of tMRCA estimate indicated that the major monophylogenetic cluster of CRF07_BC in Liangshan in Southwest China may be established approximately 13 years ago (95%PHD, 10-16 years), earlier than that in Urumqi (11 years ago; 95%PHD, [9-13]) or Yili (10 years ago; 95%PHD, [9-11]) in northwest China.
CONCLUSIONS: The epidemiology and molecular epidemiology data indicated that CRF07_BC was spread from south to north in the west part of China. This study provided direct evidence indicating such a trend that CRF07_BC in Yili may be transmitted from Liangshan or somewhere else via Urumqi through heroin trafficking route.
