BACKGROUND: The dynamic interaction between HIV and the host immune response leads to an altered expression of lymphocyte surface antigens, which reflect various aspects of immune pathology. Activation occurs early and persists throughout causing a progressive breakdown of immunity. We designed this pilot study to investigate the changes in the T-cell phenotype in HIV infected Indian patients, as there are no data available regarding the cell surface changes upon HIV- infection in the Indian population.
METHODS: Blood samples collected from 50 HIV seropositive and 20 seronegative individuals were stained for three color flow cytometry with panels of CD8, CD4, CD3, CD45RA, CD45RO, CD62L, CD38 and antiHLA-DR antibodies. Appropriate leucogate and isotype controls were included. Samples were processed by FACSLyse /wash method, acquired on a BD FACSCalibur instrument and analysed by Cell Quest software. The mean, median and SD values were determined and expression levels compared using the nonparametric Mann-Whitney test.
RESULTS: Significant increase was found in the expression of HLA-DR and CD38 on CD8 cells in HIV infected individuals. CD38+HLA-DR+ T-Cells were also significantly elevated. While there was a significant decrease in the CD45RA and CD45RO on CD4T-Cells, CD8 T-Cells showed an increase in the levels of CD45RA and CD45RO markers. Significant decrease in the CD62L +CD45RA+ on CD8+T-Cells was observed but it was not so in the CD4 subset.
CONCLUSION: Our data suggest that immune activation profile in patients infected with Indian HIV subtype C and is similar to that for other subtypes reported in previous studies. However the range of expression of these markers in the healthy population is different from that reported in Caucasian population.
