Abstract: To evaluate CD30 expression in relation to Th1 or Th2 lymphokine profile, PBL from healthy donors were activated with anti-CD3 antibodies in the presence of IL-12 or IL-4 respectively. Significant CD30 expression was observed in unsorted PBL or positively selected CD8+ cells after activation with anti-CD3 and IL-12. These lymphocytes produced a large amount of IFN-gamma, but not IL-5, therefore expressing a Th1 type of lymphokine production. IL-4 together with anti-CD3 in contrast induced PBL to secrete IL-5 but not IFN-gamma, and inhibited or had no effect on CD30 expression. The level of CD30 expression and Th profile induced by IL-12 after primary activation could be downregulated in part by IL-4 during secondary stimulation. Similarly, cells activated with IL-4 that are CD30 low can be induced to express CD30 by IL-12 in secondary activation. Costimulation with anti-CD28 antibodies also upregulated CD30 expression and lymphokine production in anti-CD3 activated lymphocytes.
