Cellular host range of human herpesvirus 8 (HHV8) (Meeting abstract).

Abstract:
Objective: To determine the in vitro tropism of HHV8. Methods: Over 40 cells and cell lines were evaluated for their susceptibility to HHV8 infection, including B cells, T cells, epithelial cells and endothelial cells (EC). Infection studies were performed by inoculation with cell-free HHV8. This virus was prepared from the body cavity-based lymphoma (BCBL) cell line, BCBL-1. Cells were inoculated with either culture fluid or sucrose-purified HHV8 from phorbol ester activated BCBL-1. The presence of HHV8 RNA was determined by reverse transcribing whole cell RNA into complementary DNA which was then used as substrate for the polymerase chain reaction (PCR) with HHV8-specific primers. Infection was also attempted by a cell contact approach in which cell lines were cocultivated with either KS biopsies or HHV8-infected cell lines derived from BCBLs. In these studies HHV8 DNA was detected by PCR. Results: HHV8 RNA was detected in inoculated primary CD19+ B cells, human umbilical vein EC and microvascular EC. HHV8 RNA could also be detected in a transformed rat liver EC line and a transformed human brain EC line. These results suggest that HHV8 had replicated in these cells. However, virus transmission studies from these HHV8-infected cells have suggested that the infection was not productive. Conclusions: The in vitro host range of the BCBL-1-derived HHV8 includes primary B cells and endothelial cells.
