Abstract: The HIV-1 regulatory gene, tat, encodes an early transactivator protein (Tat) necessary for virus replication. This Tat protein is actively secreted by infected cells. We have recently demonstrated that constitutive expression of the tat gene in a Burkitt lymphoma cell line up-regulates interleukin-4 receptors (IL-4R). The exogenous recombinant Tat protein or supernatant of Raji-tat cells also enhanced the expression of IL-4R in RCC cell lines. Because IL-13R may share component(s) with IL-4R, we investigated whether Tat protein also regulated IL-13R on RCC cell lines. We observed that Tat protein (up to 200 ng/mL) increased IL-13R expression three-fold in WS-RCC and approx 60% in PM-RCC cell line. Cycloheximide inhibited IL-13R protein synthesis in both control (t1/2 6.2 hr) and Raji-tat supernatant treated (t1/2 5.2 hr) PM-RCC cells. This Tat-mediated up-regulation of IL-13R was abrogated by either Tat-antiserum or deactivated Raji-tat supernatant indicating Tat specificity. The ability of Tat protein to modulate expression of certain cytokine receptors on cancer cells might have important implications in understanding the pathogenesis of frequent neoplastic disorders in HIV-1 infected individuals.
