High level IL-2 expression vectors using HIV LTR in human colon carcinoma cell line (Meeting abstract).

Abstract: The success of IL-2 gene therapy is dependent on high expression of the gene product. Tumor cells transduced with IL-2 and intratumoral injection of IL-2 expressing plasmids result in effective antitumor activity in some tumors. We have developed two plasmid expression vectors by replacing the CMV promoter of Promega pCl neo with HIV-1 LTR and HIV-2 LTR, respectively and cloned the IL-2 gene downstream of them. Both were further modified by the addition of another transcriptional unit expressing the Tat gene, a potent transactivator of HIV transcription. The above plasmids were transfected into human colon carcinoma cell line, SW480, using lipofection and assayed for IL-2 production 48 hours later by ELISA. The levels of IL-2 produced by HIV-1 IL-2 plasmid and HIV-2 IL-2 plasmid were comparable to the pCI neo-IL-2 (CMV promoter) plasmid, while that of HIV-1 IL-2-Tat and HIV-2 IL-2-Tat was 3 fold and 3.5 fold higher respectively. The CMV promoter is widely used to drive the expression of genes in various cell types. A possible advantage of HIV LTR-Tat promoter is that they may give higher expression than CMV in some cell types. Future studies involve transfecting different carcinoma cell lines, fresh tumor cells and T lymphocytes to observe the IL-2 production profile.
