Analysis of IL-13 receptor structure identifies three different types of receptors which utilize two different types of cloned IL-13R alpha chains (Meeting abstract).

Abstract:
We recently reported that IL-13 receptor (R) can be classified into at least three major types based on the patterns of IL-13 binding and interaction with IL-4. Recently, two different human IL-13R chains have been cloned from Caki-1 renal cell carcinoma RCC) and MT-2 HTLV-1 infected T cell lines (termed here alpha and alpha'), respectively. To investigate which IL-13R chain is utilized in each type of IL-13R, we examined various cell lines for the expression of mRNA for both IL-13R chains, IL-4R p140 and gammac chain by RT-PCR. In type I IL-13R (RCC, U251 glioblastoma and IGROV-1 ovarian carcinoma), both IL-13Ralpha and alpha' chains were expressed. In type II IL-13R users (HT-29 human colon carcinoma, A431 epidermoid carcinoma and PA-1 ovarian carcinoma), IL-13R alpha' but not alpha was expressed. In hematopoietic cells, (EBV-B cells and TF-1 myeloid cell line) IL-13R alpha' but not IL-13R alpha chain was expressed along with IL-4R p140 and gammac (type III IL-13R). However, neither IL-13R chains were detected in human T cell line, H9 and the gibbon T cell line MLA-144. These data suggest that type I IL-13R consist of a dimmer of alpha and alpha'. On the other hand, type II IL-13R consist of IL-13R alpha' and IL-4R p140 and type III IL-13R may consist of IL-13R alpha', IL-4R p140, and gammac.
