Binding antibody against primary HIV-1 isolates induced in recipients of recombinant envelope glycoprotein HIV-1 candidate vaccines (MN/rgp120 and IIIB/rgp120).

Abstract:
Induction of binding antibody to native envelope glycoproteins of primary HIV-1 isolates by MN/- and IIIB/rgp120 vaccines in alum was evaluated. A flow cytometric indirect immunofluorescence assay was used to measure the binding of serum antibodies to PBL's infected with primary HIV-1 isolates. Sera were obtained before and after vaccination of 27 normal, healthy, HIV-1 uninfected adults, who received one of four immunization regimens or placebo: 1) MN/rgp120 vaccine alone, 2) MN/rgp120 vaccine co-administered with IIIB/rgp120 vaccine, 3) IIIB/rgp120 vaccine alone, 4) priming with IIIB/rgp120 vaccine followed by boosting with MN/rgp120 vaccine, or 5) adjuvant placebo. Six clade B 92US727, 92US714, 92US657, 92US660, 91US056, BZ167), one clade A 92UG029), and one clade EA (92TH022) primary HIV-1 isolates were studied. Positive control sera from HIV-1-infected non-vaccinees who acquired HIV-1 infection in the US had high levels of antibody against all eight primary HIV-1 isolates. All 15 vaccinees who received regimens (1) and (2) made antibodies against four clade B viruses at levels half that of the infected patient sera and as high as infected patients against 91US056. Antibodies to BZ167, 92UG029 and 92TH022 were detected in sera from about 25% to 50% of vaccinees in regimens (1) and (2), but at lower levels compared to infected patients. Vaccinees who received IIIB/rgp120 vaccine alone developed lower levels of antibody against clade B viruses; these increased after MN/rgp120 vaccine boosting. The MN/rgp120 vaccine was an effective inducer of cross-reactive antibodies against envelope glycoproteins of several HIV-1 strains, and it boosted the lower antibody response induced by immunization with only IIIB/rgp120.
