Abstract: In cancer immunotherapy with biological response modifiers (BRM) such as bacillus Calmette-Guerin (BCG) or streptococcal preparation OK-432, induced interferon-gamma (IFN-gamma) plays a crucial role in the inhibition of tumor growth. This study was undertaken to determine whether additional cytokines would further enhance IFN-gamma production when added to these BRMs. IFN-gamma production by mouse splenocytes followed a parabolic dose-response curve to each BRM. When stimulated with BCG, IFN-gamma production was substantially enhanced by the addition of IL-2, IL-12, IL-15, or GM-CSF. OK-432 was a stronger IL-12 and IFN-gamma inducer than BCG. Furthermore, IFN-gamma production with OK-432 was enhanced by the addition of IL-2, IL-12, or IL-15, but not by GM-CSF. IL-10 or antibody to IL-12 markedly inhibited IFN-gamma production by both BRMs. These results suggest that both BCG and OK-432 are potent immunostimulators of the Th1 cytokine response and that this response can be accelerated by the rational addition of other cytokines.
