WSU-Br1: A novel human breast xenograft model (Meeting abstract).

Abstract: Despite treatment advances in medical oncology, metastatic breast cancer remains a serious, incurable disease taking the lives of more than 44,000 American women/year. Both new treatment modalities and new tools of drug discovery must be developed. In September, 1992, an HIV-, chemotherapy naive, 45 y/o female underwent a modified radical mastectomy for Stage IIIB, ER-/PgR-, Grade III poorly differentiated adenocarcinoma. Intra-operatively, a 1.0 cm specimen from both her primary tumor and matted nodal region were removed, cut into 30-60 mg mashed chunks and implanted into flanks of 6 SCID mice. Initial implant reached 1000 mg in a median of 78 days (d) (6/6 takes). Subsequent passages demonstrated a marked decrease in exponential doubling time (Td): 5th generation (gen) Td=4.5d; 8th gen Td=2.2d; 10th gen Td=2.1d. This model has never been passaged in tissue culture, thus avoiding the highly selective in vitro environment. It is invasive and metastatic (foci isolated in lungs in SCID mice). WSU-Br1's drug responsiveness includes: vincristine (T/C=0%; 5/5 cures (C)); taxol (T/C=0%; 2/3 C); Cytoxan (T/C=0%; 3/5 C); cisDDPt (T/C=8%; 0/4 C), VP-16 (T/C=31%; 0/5 C); Adriamycin (T/C=44%; 0/5 C); 5-FU (T/C=79%; 0/5 C); Mito-C (T/C= 101%;0/4 C). Histology remains unchanged through 10 passages. We are attempting to develop a taxol-resistant line of WSU-Br1.
