Abstract: High incidence of Kaposi's sarcoma (KS) and non-Hodgkin's lymphoma in immune-suppressed individuals suggests either a cancer inducing/promoting role of immunosuppression or the susceptibility of the immunosuppressed host to infection by communicable oncogenic viruses. We studied this question indirectly by determining the immune status of 2 strains of mice undergoing carcinogenesis by DMH, which produces both KS-like angiosarcoma and non-immune related colorectal tumors. DMH was given sc to groups of CD1 and C57BL/6 mice weekly (20 mg/kg) for 40 wk. C57BL/6 also received LPBM5, an immunosuppressive mouse AIDS virus. Animals were sacrificed at regular intervals and tested for their immune status (PFC response and spleen cell mitogenicity to LPS and Con A) and for tumor incidence by histological examination of tissues. All DMH treated mice had a 30-50 percent depression in immune responses after 20 weeks of DMH, spleen hypertrophy and mild lymphadenopathy. However, the tumor incidence was unique in each strain. Colorectal tumor/angiosarcoma ratio was 2.5 for CD1 and 0.2 for C57BL/6 and was unchanged by LPBM5. Our observations support that immunosuppression acts indirectly by increasing susceptibility of hosts to endogenous or acquired oncogenic agents.
