Abstract: We have previously reported that serum interleukin 10 (IL-10) is increased in patients (pts) with non-Hodgkin's lymphoma (NHL) and correlates to a poor prognosis. Here, we have investigated the cellular source of IL-10 and the possible relationship with EBV infection in non-HIV related NHL and Hodgkin's disease (HD). Immunostaining with 85, an anti-IL-10 antibody (Ab) recognizing both human and EBV IL-10 (vIL10) was performed in tumor samples of 37 pts with NHL and 7 pts with HD. IL-10 was detectable in 24 of 37 (65%) NHL and 5 of 7 (71%) HD. Pts with detectable serum IL-10 had consistently a positive staining for 85, suggesting that tumor cells are the source of circulating IL-10. Presence of IL-10 did not correlate to histological subtype, T/B phenotype and clinical stage. Using immunostainings with the 65 Ab (specific for vIL10), vIL-10 was detectable in 6 of 37 (16%) NHL and 1 of 7 (14%) HD pts. In situ hybridization using EBER probes, characteristic of latent EBV infection, showed that 17 NHL and 4 HD pts had EBER+ tumors. vIL-10+ tumors were consistently found EBER+ (p=0.004), whereas hIL-10 was detected in both EBER+ and EBER- NHL or HD. In conclusion, hIL-10 and/or vIL-10 are produced in vivo by malignant cells in the majority of pts with NHL and HD. Different biological mechanisms, including EBV, are involved in the deregulation of IL-10 production by NHL malignant cells.
