Abstract:
Tumor infiltrating lymphocytes (TILs) are thought to play a significant role in immune surveillance of cancer, and their presence in high numbers has been associated with better prognosis of the patients. The majority of TILs express the alpha/beta T-cell receptor (TcR), however, in some tumors gamma/delta TcR expressing TIL (greater than 4%) can be detected. In an attempt to characterize the function of 9 TILs we have screened 10 TILs bulk cultures-derived gamma/delta cytokine expanded from different malignant melanoma specimens. By FACS analysis we found in 1 out of 10 melanoma TILs cultures gamma/delta TILs which were derived from a primary cutaneous melanoma, Clark level IV. On day 34 of in vitro culture 40% of bulk TILs expressed the gamma/delta phenotype. Further analysis by double staining of FACS sorted gamma/delta TILs (purity: 99%) revealed a mixed population of 77% CD4-CD8-, 12% low CD4+, 10% low CD8+, 5% triple negative CD2-CD4-CD8- T cells and a subset of deltaTCS1+ cells. Functional activity of gamma/delta against various allogeneic tumor cells and the autologous melanoma cells displayed a strong non-MHC restricted killing and weak but specific autoreactivity. These results suggest an important role for gamma/delta T cells in immune recognition of cutaneous tumor cells.
