Abstract:
Gene therapy could be used not only to treat single gene defects, but also for many other disorders such as cancer and degenerative neurologic disease. Early attempts at gene therapy involved the indirect approach of modifying cells in culture and then transplanting them back in the body, but this book focuses on direct introduction of genes into the body without cell transplantation. Contributions from 47 authors are organized into three sections. The first provides a background for the concepts of gene therapy, with chapters on history of gene transfer, producing mouse genetic models for human diseases, post-transcriptional considerations for gene expression, and promoters, enhancers, and inducible elements for gene therapy. Section II covers methods and mechanisms with the chapter topics: mechanisms of DNA uptake in skeletal muscle; receptor-mediated gene delivery employing adenovirus-polylysine-DNA complexes; gene transfer using liposomes; in vivo receptor-mediated, and calcium phosphate-mediated DNA transfection; cellular internalization of oligodeoxynucleotides; particle bombardment--the Accell gene gun; and electrotransfection in vivo. The final section on applications covers: pharmacokinetic considerations; virus-mediated genetic treatment of rodent gliomas; gene therapy of adenosine deaminase deficiency and malignant solid tumors; herpes simplex viral vectors for the central nervous system; applications of direct and catheter-based delivery to cardiovascular diseases and malignancy; gene therapy of arthritis; gene therapy: the advent of adenovirus; gene transfer into the heart; use of asialoglycoprotein-polylysine conjugates; and retrovirus-mediated gene transfer and Duchenne muscular dystrophy.
