Assessment of lymphoid cell immunophenotype in laryngeal carcinoma (Meeting abstract).

Abstract:
Determination of cell immunophenotype of cancer patients was found useful in the assessment of immune competence. We attempted to examine an immunophenotype of laryngeal carcinoma patients at the time of surgery. We used both, patients blood lymphocytes (PBL) and tumor infiltrating lymphocytes (TIL) isolated from resected cancer tissue. TIL were isolated by mechanical and/or enzymatic disaggregation of tissue followed by centrifugation on density gradient. PBL were isolated by standard techniques. Both blood cells and TIL were allowed to react with monoclonal antibodies vs lymphocyte/monocyte differentiation antigens and analyzed by flow cytometry. It has been found that in the blood of about half of patients percent of natural killer (NK) cells (CD56+, CD16+) was increased. It was accompanied by reverse CD4:CD8 T cell ratio. When TIL were examined cell percent values did not correspond to those found in the blood of the same patient. In general, CD4:CD8 ratio was low but oscillated around 1.0. NK cell values tended to be lower while B cells (CD19+) were higher than in the blood. An interesting feature of TIL was clear-cut subset of CD3+DR+ cells. These data indicate that larynx cancer patients show alterations in lymphoid cell phenotype at time of surgery.
