Abstract: Recent European and U.S. studies comparing AZT alone or in combination with 3TC or DDC are showing the best results from a combination therapy of AZT and 3TC. Together, the studies demonstrate a clear advantage for beginning therapy with AZT plus 3TC rather than starting with AZT alone. Even people who have previously used AZT receive added benefit by switching to this combination. Data also are confirming that the AZT/3TC combination is having a longer-lasting antiviral effect than single drugs or previously-studied combinations. Unfortunately, these study findings are greatly increasing the demand for 3TC, and the sponsor can no longer meet supply needs for early access to the drug. Glaxo, the producer of 3TC, now limits enrollment conditions for the drug.
