Abstract:
We have immunized mice i.m. or intranasally and rabbits i.m. with HIV gp160 (MicroGeneSys) formulated with alum or proprietary adjuvants (PA, Pharmos) and/or meningococcal outer membrane protein proteosomes. When administered i.m., proteosomes plus alum or PA formulations (with or without proteosomes) induced improved and broadened immune responses against several gp160 epitopes compared to the standard gp160 alum-adjuvanted vaccine in both mice and rabbits. Moreover, either PA or proteosomes dramatically enhanced immunogenicity following intranasal immunization and proteosome formulation markedly boosted IgA responses. Data analyzed using a combination of ELISA, quantitative Western Blot and PEPscan techniques show that altering the presentation, formulation and route of administration of gp160 influences both quantitative and qualitative aspects of epitope specific IgG and IgA immune responses. These data are encouraging for the development of enhanced i.m. or intranasal sub-unit formulations of gp160 vaccines for HIV vaccine therapy.
