Abstract: Identification of HIV-1 sequence variants that are transmitted to infants and determination of the timing of transmission are crucial to understanding the underlying mechanisms. We analyzed fetal tissue from five abortuses and detected HIV-1 DNA in four. Analysis for maternal DNA in the fetal samples by PCR amplification of the apo-B VNTR locus detected contaminating DNA in one of the samples. The C2V3 to V4V5 region of HIV-1 envelope gene was amplified from three maternal PBMC samples and corresponding fetal tissue, inserted into plasmid vectors and sequenced. Phylogenetic analysis of the sequence variants indicated that viral sequences found in fetal tissue exhibited a low degree of divergence and were highly homologous to some of the variants in the mother. Our studies suggest that transmission could occur early in gestation.
