Susceptibility of mice with MAIDS to cytomegalovirus retinitis at times relative to Th1/Th2 shift.

Abstract:
Prior to absolute decline in CD4+ T cell number in late-stage AIDS, HIV-1 infection produces a relative immune dysregulation caused by a shift in CD4+ T helper (Th) function from a mechanism that promotes cell-mediated immunity (Th1 response) to one that favors humoral immunity (Th2 response). Theoretically, this dysregulation predisposes to opportunistic infections. Mice with retrovirus-induced immunodeficiency syndrome (MAIDS) also exhibit a Th1/Th2 shift that becomes evident approximately 3 wks after retrovirus infection. We therefore explored the susceptibility of mice with MAIDS to murine cytomegalovirus (MCMV) retinitis at times relative to Th1/Th2 shift. Groups of adult C57BL/6 mice were infected with a mixture of murine retroviruses (LP-BM5 MuLV); control animals received PBS. At 2 wks (MAIDS-2), 4 wks (MAIDS-4), and 12 wks (MAIDS-12), MuLV-infected and age-matched control mice were inoculated by subretinal injection with MCMV and compared 10 days postinfection for evidence of MCMV replication and typical retinitis. When compared with immunocompetent controls, MAIDS-4 and MAIDS-12 mice demonstrated increased ocular MCMV titers. Whereas the frequency of typical necrotizing retinitis in MAIDS-12 animals was 88%, retinitis was not observed in immunocompetent, MAIDS-2 or MAIDS-4 animals. Nonetheless, evidence of retinal infection without retinitis was observed in 0%, 12% and 70% of immunocompetent, MAIDS-2, and MAIDS-4 animals, respectively. We conclude that although typical MCMV retinitis cannot be induced at time of initial Th1/Th2 shift, increased MCMV replication and evidence of retinal infection is observed.
