Abstract: We have obtained cell cultures from biopsies of eight Kaposi's sarcoma (KS) patients. Two were derived from HIV-negative cases and six were obtained from HIV positive patients. The IST-KS cultures presented a prevalence of spindle shaped cells. Immunohistochemical staining of the cells showed expression of the smooth muscle isoform of alpha actin, the fibroblastic marker TE7 as well as the mesenchymal marker vimentin. The spindle cells were positive for desmin, but negative for the endothelial cell antigens factor VIII, EN4 and PAL-E. They grew with a sarcoma-like morphology on Matrigel. It has been reported that AIDS associated KS cell lesions give rise to angiogenic lesions in nude mice, mostly composed by the host endothelial cells (Science 242:430, 1988). Supernatants of the IST-KS cultures (KS-CM) are able to stimulate endothelial cell migration and invasion in vitro. KS-CM from three lines have been also tested in an in vivo model and they were able to induce an angiogenic response in mice. KS-CM-induced angiogenesis is inhibited by collagenase inhibitors. Antiangiogenic factors could represent potential drugs for the treatment of KS lesions.
