Abstract: It is known that some retroviruses, including HIV and SIV selectively incorporate host cell proteins into their envelopes during virus egress from the infected cell. Recent results have shown that vaccine mediated protection in macaques, seen when vaccine and challenge virus are prepared in human cells, is, at least in part, due to antibody responses directed against these host cell proteins. We have reported previously that protected macaques vaccinated with SIV grown on C8166 cells make a major antibody response to human MHC class-I antigens. No absolute correlation between such responses and protection has been seen. We now show that vaccinates also make antibody responses against HLA class-II DR antigens. These anti human responses are unique to animals vaccinated with virus grown on human cells and do not appear in infected animals. In contrast, we report that infected animals make antibody responses directed against rhesus MHC antigens. These data suggest either that SIV antigens may mimic regions of rhesus MHC molecules, in a species-specific manner, or that infected animals recognise altered or non-self MHC antigens. To determine if allo-cell responses can protect macaques against challenge with SIV grown on monkey PBMC, we have prepared and titrated an SIV cell-free challenge pool on PBMCs taken from a limited number of macaques. Four macaques were then vaccinated with paraformaldehyde-fixed monkey PBMC taken from the same animals as used to produce the virus challenge stock. Data on the immune responses and on the outcome of challenge will be presented.
