Abstract: We have previously shown that beta-2 microglobulin (beta-2M), HLA I (alpha-chain), and HLA II DR (alpha- and beta- chains) are specifically associated with purified preparations of HIV-1, HIV-2, and SIV. These cellular antigens are each present at approximately 15% of the levels of gag on a molar basis, based on recovery of these viral and cellular proteins on HPLC. Antibodies to beta-2M and HLA DR precipitated HIV-1, demonstrating that these proteins were physically attached to the viral surface. Antisera to beta-2M and HLA DR also inhibited infection of cultured cells by both HIV-1 and SIV, providing additional evidence that these cellular proteins are physically associated with these viruses and suggesting that they are involved in the infection process. Previous studies with subunit vaccines have shown that immunization with viral env antigens alone are sufficient to elicit protective immunity against SIV or HIV. The finding that macaques immunized with uninfected human cells were protected against challenge from SIV grown in human cells (E. J. Stott) raised the possibility that immune responses to cellular antigens might also be involved in protection against SIV challenge. To determine if beta-2M, HLA I and/or HLA II DR were specifically involved in this protection, we have immunized 12 M. fascicularis with these proteins and with HuT 78 cells. These macaques have developed measurable antibody titers to these proteins and are being challenged intravenously with 10 animal infectious doses (AID) of SIV/Mne grown in HuT 78 cells. These animals and unimmunized controls will be monitored for evidence of virus infection.
