Performance characteristics of HIV-2 serology reagents on blood from SIV/HIV-2 infected monkeys.

Abstract: Over the past five years, we have tested more than 20,000 sera from humans and non-human primates for antibodies to SIV/HIV-2, using first generation reagents prepared from HIV-2 viral lysate. The increasing demands for testing, and the wider commercial availability of new generation reagents have led us to evaluate the performance characteristics of various HIV-2 reagents for SIV/HIV-2 antibody detection. We evaluated assays using viral lysate (ROD, NIH-Z, GB-122), recombinant protein (env and gag) and synthetic peptide (env and gag) antigens as targets. The methods included enzyme immunoassays, Western blots, and rapid/simple tests. We also evaluated recently released combination HIV-1, 2 tests. These assays were evaluated with sera from rhesus macaques, stumptailed macaques, sooty mangabey monkeys, and African green monkeys infected naturally or experimentally with HIV-2, SIVmac, SIVsmm, SIVstm, SIVagm. All the HIV-2 reagents (viral lysate, recombinant protein, synthetic peptide) detected antibodies to all the strains of SIV tested. By Western blot, env cross-reactivity was more broadly detected than gag. Using sequential sera from animals undergoing seroconversion we hope to determine which reagents will detect antibody earliest in infection. HIV-2 reagents provide a sensitive, standardized means of detecting SIV/HIV-2 antibodies in humans and non-human primates. They are valid tools in field studies for seroprevalence/new isolates, colony screening and management, following the course of infection/disease in experimentally infected monkeys, and monitoring human exposure to infected monkeys.
