Abstract: Interleukin-6 (IL-6) is a multifunctional cytokine that functions as an autocrine growth factor for AIDS-derived Kaposi's sarcoma (KS) cells. Although some KS cells in culture have been reported to express constitutively high levels of IL-6, we find variability in the levels of IL-6 expressed by KS cells isolated from different patients (pts). In addition, IL-6 was highly inducible in all pts examined. KS cells derived from several different pts expressed low basal levels of IL-6. Incubation with either tumor necrosis factor alpha or lipopolysaccharide-induced secretion of IL-6, leading to levels that were 40- to 70-fold higher than in the uninduced state. Both IL-1 beta and the synthetic double-stranded RNA, poly (I:C), induced levels that were 100- to 200-fold above baseline. Dramatically, the combination of IL-1 beta and poly (I:C) led to a synergistic increase in IL-6 production with levels that were more than 500-fold higher than in the basal state. This suggests that in vivo situations in which KS cells encounter both IL-1 beta and double-stranded RNA would lead to high levels of IL-6 production and hence autocrine induction of KS cells. Such situations would exist in response to a number of viral infections and could explain the exacerbations of KS that occur during some systemic infections.
