Prognostic value of IL-10 in non-Hodgkin's lymphoma (Meeting abstract).

Abstract:
Serum levels of interleukin 10 (IL-10) were measured retrospectively in the serum of 60 normal subjects and 154 patients (pts) with non-Hodgkin's lymphoma (NHL), including 102 with evolutive NHL (87 at diagnosis, 15 in relapse) and 52 in first remission. We used an ELISA measuring both human IL-10 (hIL-10) and viral IL-10 (vIL-10, ie, the EBV BCRF1 molecule). IL-10 was detectable in 48 (47%) of 102 pts with evolutive NHL and on 3 of 52 (6%) pts in first remission (p less than 10(-4)). IL-10 was detectable with a similar frequency in all histological subtypes of NHL. Among the 70 pts with intermediate or high grade NHL, those with detectable serum IL-10 at diagnosis had a significantly shorter overall (p = 0.025) and progression free (p = 0.030) survival. Serum IL-10 was associated with a very poor prognosis in pts (p = 0.00004). Multivariate analysis with Cox model indicate that IL-10 is an independent prognosis factor for both overall and progression free survival in these pts. Using a second ELISA specific for vIL-10, IL-10 was identified as vIL-10 in 23 of the 48 pts with evolutive NHL and in none of the 11 EBV seronegative and in 30% of EBV seropositive pts. vIL-10 was detectable only in the serum of pts with B lymphoma (p = 0.03). These results strongly suggest that both hIL-10 and vIL-10 are involved in the physiopathology of non-HIV related NHL.
