Abstract: Angiogenesis and in some cases edema are two major components of AIDS-KS. Cultured spindle cells isolated from various KS lesions of patients with AIDS transplanted in nude mice induce KS-like lesions of murine origin. The critical factor for this purpose is oncostatin H. We report here that AIDS-KS cells also release another factor that mediates vascular hyperpermeability (VHP) resulting in edema in our model system. This factor in CH induces VHP and growth of vascular endothelial cells. These activities bind heparin sepharose and elute in 0.5 M NaCl fraction. The endothelial cell growth promoting activities were identified in 0.5 M and 2.0 M NaCl fractions. Anti-bFGF IgG inhibited endothelial cell growth promoting activity in 2.0 M NaCl fraction. Anti-VEGF serum inhibited VHP and endothelial cell growth promoting activities of 0.5 M NaCl fraction. Both 0.5 M and 2.0 M NaCl fractions induced angiogenesis in rabbit cornea assay. Significant level of VEGF-specific mRNA was present in AIDS-KS cells (3.9 kb band). In summary, it appears that AIDS-KS cells produce VEGF and bFGF-like activities that mediate two important features of AIDS-KS: edema and angiogenesis.
