Abstract: Recent interest has focused on the different biological properties of HIV-1 isolates. Non-syncytium-inducing (NSI) isolates, detectable at all stages of HIV infection, are preferentially monocytotropic in contrast to SI isolates which precede symptoms of AIDS and are T-cell tropic. We are examining the effects of NSI and SI virus strains on the proliferative responses of seronegative PBMC in vitro. Peripheral lymphocytes from seronegative donors were infected with NSI and SI virus at similar MOI, and PHA, autologous EBV LCL and tetanus toxin were added at 0, 3, 7 and 14 days postinfection (PI). At 3, 7 or 14 days responsiveness to PHA was reduced by as much as 56-66% in the cells of donor WC infected with the SI isolate compared to the response of cells infected with the NSI isolate. The responses to EBV antigen were also lower in cells infected with the SI isolate at 3, 7 or 14 days PI. This observation is being further studied to determine conclusively whether SI virus interferes with proliferative responsiveness in vitro.
