Abstract: Tumor onset is significantly delayed (21 days later than controls) by feeding a folate restricted diet to mice transgenic for the human T-lymphotropic virus type 1 tax (HTLV-1 tax) gene (J Natl Canc Inst 84(5):332-7, 1992). These mice develop tumors with a well-defined time of onset and tissue involvement. To test other dietary interventions known to modulate tumorigenesis, 16 female mice heterozygous for the HTLV-1 tax construct were bred to CD-1 males and 60 of their offspring identified as transgenic (heterozygous) for the HTLV-1 tax gene construct by PCR were systematically assigned at weaning to either a control, a calorie-restricted (80% of control) or a high-fat (25% fat vs 5% for control) diet using litter and gender as blocking factors. Mice were carefully examined at least 3x/wk for visible tumors. The mean time to first tumor onset was 82.9 +/- 5.5, 92.9 +/- 6.5 and 89.4 +/- 7.0 days (p=0.56, ANOVA) for mice fed control, calorie-restricted or high-fat diets, respectively. Median time to tumor onset was estimated to be 85 days. The overall p-value for the Wilcoxon test was p=0.52, group comparisons were p=0.20 for control vs restricted, p=0.79 for control vs high-fat and p=0.50 for restricted vs high-fat. No effects of gender or litter were detected. Thus, although mice fed calorie-restricted diets had an approx 10 days delay in tumor onset, this effect was not significant with this sample size and it is concluded that there is a unique effect of folate restriction in this mouse model of tumorigenesis. Future studies will focus on molecular and biochemical mechanisms of nutritional effects on tumor onset in this and other transgenic models of tumorigenesis.
