Recognition of a mutated region of the surface glycoprotein of HTLV-I by human sera (Meeting abstract).

Abstract:
The region comprised between amino acids 175 and 199 of the HTLV-I envelope glycoprotein is known to be one of the immunodominant domains of this molecule (Horal et al, 1991). This region is recognized by several monoclonal antibodies (MAbs): 0.5 alpha (Matsushita et al, 1986), LAT-27 (Tanaka et al, 1991), KE36-11 (Kuroki et al, 1992) and 7G5D8 (Londos-Gagliardi et al, 1993). In some HTLV-I isolates, the proline at position 192 is substituted by a serine (Malik et al, 1988). One may predict that this mutation modifies the secondary structure of the 189-199 region. To study the inference of the presence of proline or serine at position 192 on the human immune response, four 10-mer peptides overlapping in the region 186-199 were synthesized: 186-Pro-195 and 186-Ser-195 (corresponding to the epitopes of MAbs 0.5 alpha, KE36-11, and 7G5D8), 190-Pro-199 and 190-Ser-199 (corresponding to the epitope of LAT-27). Serum samples from HTLV-I seropositive patients were assayed for their reactivity with these synthetic peptides by enzyme-linked-immunosorbent assays. The percentages of reactive sera with each synthetic peptide were the following: 91% (50/55) for 175-Pro-199, 5% (3/55) for 186-Pro-195, 42% (23/55) for 186-Ser-195, 56% (31/55) for 190-Pro-199 and 62% (34/55) for 190-Ser-199. These results confirm that the 175-199 region is well recognized and that the internal region 190-199 is more often recognized than the 186-195 region by antibodies of HTLV-I infected subjects. It was observed that Ser in place of Pro at position 192, increased the reactivity of numerous sera. In addition a differential recognition of the 190-Pro-199 and 190-Ser-199 was shown, indeed the two peptides were detected, respectively, by 100% (15/15) and 73% (11/15) sera of patients with neurological disorders, 47% (9/18) and 72% (13/18) sera of lymphomatous patients (ATL) and 32% (7/22) and 45% (10/22) sera of asymptomatic HTLV-I carriers (AC). Moreover, 7 sera (4 from ATL patients and 3 from AC) reacted only with 190-Ser-199.
