Abstract: Forty-eight patients (pts) with AIDS-KS refractory to or intolerant of conventional systemic chemotherapy were evaluated for response to D-SL. D-SL 20 mg/m2 was administered every 3 wk (with cycle length modified depending on adverse events and response) for up to 10 cycles. KS lesions were evaluated according to ACTG criteria prior to trial entry, prior to and at the end of every cycle, and at 4 weeks following the end of the last cycle. Pts with partial response (PR) were followed every 2 months to determine response duration. The mean CD4 count of the study population was less than 50/mm3. Results are shown in a table. Mean cycle number at BR was approx 2.7 and the mean duration of PR was approx 83 days. D-SL was well tolerated. The majority of adverse events reported were mild or moderate in severity. Conclusions: (1) D-SL can produce PRs in up to two thirds of pts with AIDS-KS refractory to conventional chemotherapy; (2) PRs may be prolonged; (3) BRs are generally produced within the first several treatment cycles; (4) D-SL is generally well-tolerated with acceptable toxicity.
