Abstract: B-cell lymphomas are a major cause of death in long-term AIDS survivors. The NCI has established an in vitro subpanel of human lymphoma lines to identify new chemotherapeutic agents with antilymphoma selectivity. Agents referred from the NCI in vitro anticancer drug screen are tested in vitro against 10 human B-cell lymphoma lines using a new propidium iodide fluorescence assay to quantitate cellular responsiveness. Over 450 agents have been evaluated to date. Lymphoma-selective agents are tested for in vitro growth recovery over a 60-day period, then evaluated in vivo against xenografts grown in SCID mice. Lead compounds have emerged with novel chemical structures.
