Abstract: Mycoplasma penetrans, a new species of human mycoplasma isolated from patients with AIDS, has a unique tip-like structure and displays prominent pathobiological properties associated with virulence. All 6 clinical isolates share extensive antigenic similarity and DNA homology. We have studied serial serum samples from 5 of the 6 patients for M. penetrans-specific antibodies by ELISA and Western blot analysis using lipid-associated membrane proteins (LAMPS) as antigens. Four patients seroconverted and produced prominent antibodies to M. penetrans at the time of mycoplasma isolation. However, one mycoplasma-infected patient failed to mount detectable response of antibodies to this microbe. The positive antibody reactivity was mainly directed against the two major LAMPS of M. penetrans, with molecular mass of 35 and 38 kDal, respectively. Documentation of seroconversion in serial serum samples from patients known to be infected with M. penetrans further validates our serological assays for mycoplasma-specific antibodies using LAMPS.
