T cell mitogenesis-inhibitory activity of phospholipid fraction of M. intracellulare.

Abstract: Previously, phospholipid (PL) and polar mycoside fractions of M. avium complex (MAC) were found to have immunosuppressive activity against ConA-induced T cell mitogenesis. In this study, we investigated the immunosuppressive mechanism of action of the PL fraction, which was prepared by serial extractions with acetone, hot methanol, and hexane from lyophilized cells of M. intracellulare N-260 strain. When murine spleen cells (SPCs) were cultivated in RPMI medium containing 2 micrograms/ml of ConA in the presence of the PL fraction (62.5-250 micrograms/ml), ConA-induced IL-2 production and the subsequent proliferative response of T cells was markedly reduced. Although T cell activation to IL-2-reactive state was also inhibited by the PL fraction, it was not inhibited as much as IL-2 production. Authentic PLs, such as phosphatidylinositol (PI) and phosphatidylserine (PS), but not phosphatidylcholine or phosphatidylethanolamine, suppressed ConA-induced SPC mitogenesis. PI and PS caused equal reductions in both IL-2 production and generation of IL-2-reactive T cells. The MAC PL fraction, PI, and PS inhibited mitogenesis in an irreversible fashion but phase dependency of their action differed from each other. Therefore, there are some differences between MAC PL and authentic PLs in the mode of their inhibitory actions against SPC mitogenesis.
