Physical mapping of nine genes to the chromosome of rat Pneumocystis carinii.

Abstract:
Despite being an important AIDS-related pathogen, little is known about the genome of Pneumocystis carinii. The identification of essential housekeeping genes and comparison of the genes to other organisms will aid in the phylogenetic determination of P. carinii as well as identify potential therapeutic targets for improving the treatment of P. carinii pneumonia. The current study describes the isolation and identification of nine highly conserved genes from rat Pneumocystis carinii. This was accomplished by PCR amplification with degenerate primers to conserved domains and DNA sequencing. These genes are DNA polymerase II, beta-tubulin, RNA polymerase I, RNA polymerase II, RNA polymerase III, calmodulin, an alpha-like actin, a centractin-like actin, and topoisomerase II. The nine genes have been mapped to seven different chromosomes by CHEF gel analysis with three of the genes, beta-tubulin, alpha-actin and RNA polymerase I, mapping to a 440 kilobase chromosome. Preliminary studies are also described that investigate the differences among these genes from rat Pneumocystis carinii isolates and organisms isolated from human samples.
