Abstract: Over a two year period, 25 adenovirus (AV) isolates were recovered from fecal samples of 14 HIV-seropositive males. Specimens were inoculated into A-549 cells. Virus replication as evidenced by cytopathic effect was confirmed by specific AV immunofluorescent staining. Using conventional serum neutralization (SN) three isolates were identified using antisera against AV serotypes 1-39; 14 were identified using antisera against the new candidate strains, 42-47; and eight could not be identified. To further characterize the AD isolates, restriction endonuclease digestion of viral DNA was performed. Viral DNA was extracted from infected cells using a modification of the Hirt procedure, purified, concentrated and then subjected to digestion with restriction endonucleases, HIND III, BAM HI and Sma I. Restriction fragments separated by agarose gel electrophoresis were stained with ethidium bromide and viewed under UV-illumination. Analysis of the gel patterns confirmed the SN results and demonstrated that isolates obtained from the same patients had identical genotypes, and that the genotypes of isolates varied among different patients. Four unique restriction patterns were found among the eight non-serotypable viral isolates. These may represent new AV enteric pathogens associated with immunosuppressed patients.
