Abstract: HPV infection is associated with cervical cancer, and HIV appears to be a cofactor that accelerates its development. This study demonstrates that HIV-1 can infect cervical epithelial cells and synergize transactivation induced by HIV-1 tat and molecular clones of human herpesvirus-6 (HHV-6), pZVH14 and pZVB70, which are, per se, weak transactivators of HIV-1 LTR CAT. Positive infection occurred with HIV-1BaL (originally isolated from primary cultures of lung) but not with HIV-1(IIIB) (lymphotropic virus). Cervical cells lack the CD4 receptor; OKT4a antibody did not block infection. ELISA analysis of HIV-1 p24 core antigen demonstrated production of HIV-1 virus. These results, including identification of potential HIV transactivators which synergize with Tat in 2 genomic clones of HHV-6, may be important for the dissection of the molecular mechanisms of interaction between viruses in human genital cells.
