Molecular characterization of STLV-1-positive lymphomas in baboons (Papio hamadryas).

Abstract: Epizootiology, pathology, virology and immunology of malignant lymphomas in baboons (Papio hamadryas) were described earlier (for review see B. Lapin in: Viral Diseases in Laboratory and Captive Animals, Ed. G. Darai, 1988, p. 135). A strong, presumably aetiological linkage was found between these lymphomas and the HTLV-1-related virus--STLV-1bab (A. Voevodin et al., Int. J. Cancer, 36:579, 1985; Modern Trends in Human Leukemia, 31:392, 1987). The following results of our subsequent studies will be presented: all 30 baboon lymphomas tested were positive for STLV-1bab by Southern blot analysis and STLV/HTLV-1 specific PCRs; one or two STLV-1bab proviruses were integrated in individual site in each baboon lymphoma; based on restriction fragment patterns revealed by purified HTLV-1 probes under stringent hybridization conditions STLV-1bab is similar, but clearly different from prototype HTLV-1. Polymorphism of STLV-1bab proviruses integrated in baboon lymphomas was also found; tax/rex fragment amplified by PCR (A. Voevodin et al., Nonhuman Primate Models for AIDS, Seattle, p. 101, 1991) from baboon lymphoma was sequenced (118bp without primers). The homology with corresponding genomic regions of different HTLV-1s was 94-95%, pigtailed macaque STLV-1--91%. Sequencing of other genomic regions is in progress. Non of 26 baboon lymphomas was positive for B-monoclonality as determined by PCR for VDJ of rearranged IgH; monoclonal rearrangement of TCR beta C gene was found in some, but not all baboon lymphomas. Sera of both lymphomatous (61) and healthy (63) STLV-1bab positive baboons gave similar western blot patterns when tested against prototype HTLV-1. None of 20 baboon lymphomas tested by generic SIV PCR was positive. Molecular taxonomy of STLV-1s and their use as models of HTLV-1 associated diseases will be reviewed and discussed.
