Abstract: A quantitative virus isolation assay was developed for the determination of virus load in HIV-1 infected chimpanzees. In a study designed to study immunological correlates of HIV-1 vaccine protection, two different recombinant HIV-1 envelope preparations were used. Recombinant forms of HIV-1 env gp160 and gp120 (III B isolate, BH 10 molecular clone) were produced in mammalian and insect cells respectively. The purified recombinant proteins were formulated in an oil in water emulsion vehicle with 3D-MPL immunoadjuvant. Group of 2 chimpanzees were injected with 4 doses of 100 micrograms of gp160 or gp120 or a control antigen in the same adjuvant over an eight month period. Prechallenge immunological work-up included ELISA evaluation of antibody response to the immunogen, to the BH 10 V3 loop, gp120 sCD4 binding interference assay, as well as type- and group-specific neutralizing activity. DTH responses were monitored after the second boost. Intravenous homologous challenge was performed with 10 CID50 of cell-free HIV-1 III B stock. Post-challenge follow-up included prechallenge serological assays plus quantitative virus isolation, Western blot, PCR of PBMCs, lymph node and bone marrow from protected animals. Results one year post-challenge will be presented.
