Modification of viral load after immune stimulation of SIV-infected rhesus monkeys.

Abstract:
OBJECTIVES: To assess the influence of adjuvant injection on the clinical status and on the seric and PBMC viral loads of SIV infected monkeys. METHODS: Two groups of two rhesus monkeys were injected intravenously with cell free SIVmac251 or SIVsm, respectively three and one year before they were injected with 1 ml of Incomplete Freund Adjuvant (IFA) alone by the intramuscular route. Seric viremia were preformed in 96 culture microplates: three-fold dilutions of monkeys' sera (in triplicate) were incubated with 25.10(3) CEMX174 cells for 5 to 7 days; titers were determined as the last dilution where at least one out of three wells exhibited syncytia formation. Cellular viremia were performed in 96 culture microplates: two-fold serial dilutions of fresh or frozen monkeys' PBMC were cocultured with 150.10(3) CEMX174 cells; titers were determined as described above. Cytopathic effects observed were confirmed by specific anti-SIV ELISA. RESULTS: 1) Before IFA injection: no major modification of the clinical status of the four monkeys was detected, except a generalized and persistent lymphadenopathy since day 15 post infection; all animals' sera exhibited high and persistent anti-SIV antibody titers as anti-SIV neutralizing activities; proviral DNA was detected by PCR in the PBMC of the four animals. Viremics titres (weekly tested) remained stable during 40 days before the adjuvant injection. 2) Two to five days after IFA injection, all the monkeys exhibited a transient increase of their peripheral neutrophil counts, and a transient decrease of their circulating thrombocytes and lymphocytes. One out of the four monkeys developed a transient increase of its cellular viremic titer (about one log10 at day 2 post injection) which reached its former levels three weeks later.
