Passive immunization and challenge of rhesus macaques with SIVMAC.

Abstract: 14 monkeys were infused with purified immunoglobulin (Ig) or heat inactivated plasma prior to inoculation with 10-100 I.D. 50 of cell-free SIVMAC. 4 of 8 animals infused with plasma from an SIV-immunized animal were protected against infection. None of the 6 animals that were infused with plasma or Ig from infected animals were protected, despite the fact that plasma from infected animals consistently had higher neutralization titers than plasma from infected animals. The protocol for infusion and SIV challenge was as follows: Group 1.) 2 animals infused I.V. with 400 mg/kg. of Ig purified from a pool of plasma from SIV-positive, healthy animals. The recipients were inoculated I.V. with 10-100 I.D. 50 of SIV 4 hrs. post-infusion. The SIV-neutralization titer of the Ig pool against the challenge stock was 1:512 Group 2.) 4 animals infused with 10 ml/kg of immune-plasma from a single SIV-immunized animal. The recipients were inoculated with virus at 4 hours post-infusion. The SIV-neutralization titer of this pool was 1:40. Group 3.) 25 ml/kg of immune-plasma from the same pool as group 2. These animals were inoculated at 18 hours post-infusion. Group 4.) 25ml/kg of immune/infected-plasma from a single infected donor. The recipients were inoculated at 18 hours post-infusion. The SIV-neutralization titer of this pool was 1:1024. All reagents used for infusion were heat-inactivated for 30 min. at 56 degrees C. Despite development of antibody titers that were comparable to the original pool, all animals from groups 1 and 4 were found to be culture positive by 1 week post-inoculation (PI). 3/4 animals in group 2 and 1/4 animals in group 3 were found to be culture positive by 1 week PI. The uninfected animals have remained virus-negative for more than 19 weeks. They have been tested for virus by cocultivation with human PBMC and PCR. In all 4 uninfected animals SIV-specific antibody decreases with time.
