Abstract: Four pregnant female rhesus monkeys were immunized with three 100 micrograms doses of formalin-inactivated SIV + SAF-M during pregnancy, with the final immunization given 2-4 weeks prior to delivery. One infant was born prematurely due to an incompetent cervix in the mother and died. The remaining 3 infants were delivered vaginally at term and were taken from their mothers prior to breast feeding and nursery-reared. Paired mother:infant serum taken on the day of birth showed identical antibody profiles by western blotting. One to 4 days after birth, each infant was inoculated intravenously with 10 animal infectious doses of SIV/DeltaB670. One naive control monkey was similarly inoculated immediately following each infant. All 3 controls became infected as evidenced by detection of SIV-specific LTR and pol sequences by PCR in peripheral blood lymphocytes. In contrast, none of the infants born of immunized mothers has shown signs virus infection post inoculation. These data show that immunization with an inactivated whole SIV vaccine during pregnancy causes no apparent deleterious effects on neonatal development and is highly effective in protecting infants exposed to SIV at birth.
