Abstract: Cynomolgus macaques were infected with SIV or vaccinated with one of three different recombinant envelope proteins. Their antibody responses were quantitated by ELISA using, as antigens, native virus, recombinant proteins or overlapping peptides analogous to the entire envelope protein. Plasma were also examined for antibody which competed with the binding of monoclonal antibodies to conformational neutralizing epitopes on SIV gp120. All animals had high titres of antibody to SIV envelope. However, significant differences were observed in the fine specificity of antibody responses to infection and to individual recombinant proteins. In general, vaccinated animals reacted to more peptides of gp120 than infected animals. The converse was true with peptides of gp41. Most animals responded to peptides in the first and third variable regions of gp120 and to a peptide analogous to the Gnann epitope of gp41. The main differences in reactivity were in the first, second, third and fifth constant regions and in the second variable region of gp120. Antibody from infected macaques also inhibited the binding of monoclonal antibodies to conformational neutralizing epitopes of SIV.
