Abstract: We have previously demonstrated that a variety of human solid tumors express high-affinity IL-4R, and IL-4 can inhibit growth of these cells in vitro. In the present study, we have examined (1) the expression of IL-4R on AIDS-KS-derived spindle cells at the tumor cell surface and gene levels and (2) the effect of IL-4 on KS cell growth and regulation of Oncostatin-M and IL-6 expression. AIDS-KS cells expressed 1828 +/- 408 (mean +/- sem) high-affinity IL-4R/cell, with a Kd of 154 +/- 37 pM (mean +/- sem) and expressed in a single 4-kb species of mRNA for IL-4R. IL-4 inhibited KS cell proliferation in vitro in a dose-dependent manner, and this effect was neutralized by anti-IL-4 antibody. IL-4 also decreased the level of immunoreactive IL-6 and steady-state mRNA levels for IL-6 and Oncostatin-M, both of which act as autocrine growth factors for AIDS-KS-derived spindle cells. These results suggest that IL-4 could have therapeutic efficacy in the treatment of AIDS-KS, and the growth-inhibitory effects of IL-4 may be mediated through the inhibition of IL-6 and Oncostatin-M production.
