Abstract: The human immunosuppressive virus (HIV) and simian immunosuppressive virus (SIV) possess surprisingly complex genomes. In addition to three structural genes, gag, pol and env, which commonly are found in replication-competent retroviruses, HIV-1 has at least six regulatory genes in its genome. Two of these genes, tat and rev, are essential for virus replication. The remaining genes (nef, vif, vpu and vpr) are not required for virus replication, but mutations in these genes alter the replication properties of the virus. Studies of these latter genes are reviewed, including structure and function of human and simian immunosuppressive virus nef proteins (conservation of the nef genes among lentiviruses, cellular localization of nef, expression of myristoylated nef in bacteria, function of HIV and SIV nef proteins and relevance of in vitro and in vivo activities of nef); the vpr protein of HIV-1 (a virion-associated positive regulatory protein); and the nef gene of HIV-1 (recent results).
