Abstract: Until recently, the occurrence of human T-lymphotropic virus type I (HTLV-I) infection and disease was controversial in India. We have recently identified cases of HTLV-I-associated adult T-cell leukemia and spastic paraparesis among residents of south India. Our serological surveys have also uncovered HTLV-I carriers and 2 individuals dually infected with HTLV-I and HIV-1. To clarify the genomic heterogeneity of HTLV-I strains from south India, we amplified selected regions of the gag, pol and env genes in DNA extracted from whole blood blotted onto filter paper. Sequence analysis and comparison of these HTLV-I strains with those of cosmopolitan strains of HTLV-I revealed that the HTLV-I strains from south India were 98.9% similar to the Japanese prototype HTLV-I ATK. All base substitutions were silent, and no frame shifts or disease-specific base changes were found. Dendrograms based on 1287 bases indicated that the south Indian HTLV-I strains clustered with those from Japan, suggesting a common origin or source with subsequent parallel evolution. Further molecular genetic studies of HTLV-I and HIV from India, now underway, may provide additional insights into the early and recent dissemination of these retroviruses.
