Abstract: Primary cultures of human submucosal mesenchymal (SM) cells from the intestine infected with the human immunodeficiency virus (HIV) displayed Kaposi's sarcoma (KS)-like changes in growth and morphology. More IL-6 was released from HIV-infected than control cultures at early times (3-6 days postinfection) even though virus replication did not occur. In long-term studies (weeks to months) persistent or latent virus was sometimes detected, as were shifts in specific markers of angioblast/endothelial cells. Angiogenesis studies of culture supernatants were positive. Similar effects on cell growth and cytokine induction were found upon transfection of the HIV tat gene into the SM cells. This is a good in vitro model that demonstrates cytokine induction of a KS phenotype, differentiation of endothelial and mesenchymal cells, and angiogenesis.
